In Situ hybridization is the technique to use when you want to localize a specific mRNA or DNA species in tissue, cell or chromosomes.

Isotope labeled

 

Digoxygenin labeled

Isotope labeled probe such as S32 are commonly used when very high sensitivity is needed due to low copy number of target RNA in the observed tissue.   An good alternative to isotope labeled probe is digoxygenin labeled. The great advantage by not using radioactive material comes with a slight lower sensitivity.
